An effective system for isolation of mesophyll protoplasts was developed. The effect of several enzyme mixtures was tested. An yield of 20.4 x106 protoplsts/g leaf tissue with viability 92% was obtained after digestion of tissue in enzyme solution containing cellulase "Onozuca" RS – 1%, hemicellulase – 1%, pectolyase Y-23 – 0.1% and 0.7M manitol as osmotic.
The plating of protoplsts in medium containing MS basal medium with NAA – 0.01 mg/l, BAP – 1.5 mg/l and zeatine – 0.5 mg/l with osmotic as 0.5M sucrose had better influence on the beginning of first division and development of protoplasts.
